I could never really make my brain go there with Wincest. It's no offense to anyone who sees it. I'll admit during a couple of the early seasons there were scenes and moments that I thought had far more of a romantic undertone than a familial one. But in general, I never get that vibe from them. Yes, their relationship is dysfunctional and co-dependent, and they have a deep love for each other, but I don't believe what they feel is sexual or romantic in any way.
I just am not really an incest fan. I've read a few Merlin/Arthur AUs with incest. But usually it helps that I know in canon they're not brothers and usually they're either half-blood relatives and/or there's some kind of reincarnation angle. That's the only way I can really deal with incest in fiction.
It's not so odd since this is a show without many canon couples, but it's a show that I've never really rooted for any couples, slash or otherwise. I like Dean and Cas' friendship, but I just don't see either longing for the other romantically, and I don't react to their chemistry that way.
